2-Amino-4-morpholinopyridine is a useful reactant for organic reactions and synthesis. | [Synthesis]
Example 1: 4-Chloropyridin-2-amine (800 mg, 6.22 mmol) was mixed with morpholine (8.0 mL) in N,N-dimethylacetamide (DMA, 2 mL) in a sealed vessel. The reaction mixture was placed in a microwave reactor (CSA Discover) and heated at 20 °C for 5 min. After completion of the reaction, it was cooled to room temperature and the reaction mixture was concentrated under reduced pressure to remove the solvent. The crude product was purified by silica gel column chromatography with an eluent ratio of dichloromethane/methanol/ammonia (95:5:0.5, v/v/v) to afford 837 mg of the target compound 2-amino-4-(4-morpholinyl)pyridine as a yellow solid in 75% yield. The structure of the product was confirmed by 1H NMR (DMSO-d6): δ 7.70-7.64 (m, 1H), 6.59-6.54 (m, 1H), 6.05-6.00 (m, 1H), 3.74-3.65 (m, 4H), 3.45-3.37 (m, 4H). Mass spectrometry analysis showed that the measured value of [M+H]+ was 180.11 (calculated value: 179.20).
