合成4-氯噻唑-5-甲醛(333)的一般步驟:在0°C下,向攪拌的4-氯-5-(1,3-二氧戊環(huán)-2-基)噻唑(332,1.1g,5.75mmol)的THF(10mL)溶液中緩慢加入5N HCl水溶液(6mL)。將反應(yīng)混合物逐漸升溫至室溫并持續(xù)攪拌3小時。反應(yīng)進程通過TLC監(jiān)控。反應(yīng)完成后,將混合物倒入鹽水(20mL)中,用EtOAc(2×50mL)萃取。合并有機相,用飽和碳酸氫鈉溶液(100mL)洗滌,經(jīng)無水硫酸鈉干燥,過濾后減壓濃縮,得到目標產(chǎn)物4-氯噻唑-5-甲醛(333,800mg,95%),呈黃色油狀。TLC條件:10% EtOAc/己烷(Rf=0.3);1H NMR(CDCl3,500MHz):δ 10.11(s,1H),9.00(s,1H)。
