在0℃及氮?dú)獗Wo(hù)下,向攪拌的1-(2,4-二甲基苯基)-2-[(3-甲氧基苯基)亞甲基]酰肼(500mg,1.97mmol)和三乙胺(0.33mL,2.36mmol)的二氯甲烷(5mL)溶液中緩慢滴加三氟乙酸酐(0.33mL,2.36mmol)。反應(yīng)混合物在0℃下持續(xù)攪拌1.5小時(shí)。反應(yīng)完成后,將混合物濃縮,殘余物通過(guò)柱色譜法使用丙酮/己烷(1:10)作為洗脫劑進(jìn)行純化,得到目標(biāo)產(chǎn)物2,2,2-三氟乙酸1-(2,4-二甲基苯基)-2-[(3-甲氧基苯基)亞甲基]酰肼(571mg,收率83%),為淺黃色油狀物。1H NMR(CDCl3)數(shù)據(jù)如下:δ7.26-7.23(m,4H),7.19(d,J = 8.0Hz,1H),7.12(d,J = 7.5Hz,1H),7.03(d,J = 8.0Hz,1H),6.92(dd,J = 8.5,2.0Hz,1H),3.79(s,3H),2.39(s,3H),2.08(s,3H)。高分辨質(zhì)譜(ESI-TOF,m/z)分析結(jié)果:C18H18N2O2F3([M + H]+)的計(jì)算值為351.1320;實(shí)測(cè)值為351.1311。

Result: | While both young and old animals recognized when an object was moved a large distance (135 degrees), a reduction in the recognition index (RI) in aged mice was observed when the object was moved a smaller distance of 45 degrees. The reduction in the RI was reversed upon treatment with J147. |
