在500 mL圓底燒瓶中,將4-(6-硝基吡啶-3-基)-5,6-二氫吡啶-1(2H)-羧酸叔丁酯(4.9 g,16.0 mmol,1.00當量)溶于乙醇(300 mL)和乙酸乙酯(75 mL)的混合溶劑中,加入鈀碳(1.32 g,1.24 mmol,0.0773當量)。將反應(yīng)混合物用氫氣置換兩次,然后在氫氣氛圍下(使用氫氣氣球)攪拌過夜。通過LC/MS分析確認反應(yīng)完成。反應(yīng)混合物用氮氣吹掃后,通過硅藻土墊過濾,硅藻土墊用乙酸乙酯洗滌數(shù)次。合并無色濾液和洗滌液,加入二氯甲烷,將溶液蒸發(fā)至干。再次加入二氯甲烷,溶液經(jīng)真空濃縮,得到目標產(chǎn)物4-(6-氨基吡啶-3-基)哌啶-1-甲酸叔丁酯,收率定量。(M + H)+ = 278 m/z。
