以2,4-二羥基苯甲醛為原料合成5-氯-2,4-二羥基苯甲醛的一般步驟:在0℃及氬氣保護(hù)下,向2,4-二羥基苯甲醛(20g,144.92mmol)的乙醚(500mL)溶液中緩慢滴加磺酰氯(14mL,171.2mmol)。滴加完畢后,將反應(yīng)混合物升溫至室溫并繼續(xù)攪拌30分鐘。隨后,將反應(yīng)混合物倒入冰水中,分離有機(jī)層。有機(jī)層依次用水和飽和食鹽水洗滌,無水硫酸鈉干燥,過濾后減壓濃縮,得到粗產(chǎn)物。粗產(chǎn)物通過硅膠柱層析純化(洗脫劑:石油醚/乙酸乙酯=90:10),得到5-氯-2,4-二羥基苯甲醛,為灰白色固體(6.7g,收率26%,LC/MS純度97%)。ESI-MS, m/z 170.9 [M-H]?;元素分析符合C7H5ClO3;1H NMR (DMSO-d6, 400 MHz): δ 11.39 (1H, s), 10.87 (1H, s), 9.98 (1H, s), 7.0 (1H, s), 6.58 (1H, s)。
