以4-溴-5-氟-2-硝基苯胺為原料合成4-溴-5-氟苯-1,2-二胺的一般步驟:向4-溴-5-氟-2-硝基苯胺(1.0g,4.3mmol)的THF(9.0mL)溶液中加入EtOH(9.0mL)和H2O(3mL),隨后加入鐵粉(1.2g,21.3mmol)和氯化銨(0.34g,6.4mmol)。將反應(yīng)混合物在95℃下加熱回流4小時。反應(yīng)完成后,將混合物冷卻至室溫,用EtOH稀釋,通過硅藻土墊過濾,直至濾液無色。濾液經(jīng)減壓濃縮后,將殘余物溶解于EtOAc中,依次用H2O和鹽水洗滌,有機相用無水Na2SO4干燥,過濾并濃縮。向濃縮物中加入己烷,過濾收集析出的固體,得到標題化合物4-溴-5-氟苯-1,2-二胺(710mg,3.5mmol,收率81%)。
