以(S)-2-((((9H-芴-9-基)甲氧基)羰基)氨基)-5-氨基-5-氧代戊酸(Fmoc-Gln-OH)為原料合成Fmoc-L-2,4-二氨基丁酸(Fmoc-Dab-OH)的一般步驟如下: A. 將100 g(271.5 mmol)Fmoc-Gln-OH懸浮于2 L由乙酸乙酯、乙腈和水按體積比2:1:1組成的混合溶劑中。在20-30℃條件下,緩慢加入105.1 g(325.9 mmol)二異丙基乙胺(DIPA),反應(yīng)持續(xù)72小時(shí)。反應(yīng)完成后,經(jīng)過后處理得到80 g Fmoc-Dab-OH,產(chǎn)率為86.5%。產(chǎn)物經(jīng)高效液相色譜(HPLC)分析顯示純度為99.6%,并通過紅外光譜(IR)和核磁共振(NMR)確認(rèn)了其結(jié)構(gòu)正確性,相關(guān)譜圖見圖1和圖2。
