6-羥基煙酸乙酯的制備:向6-羥基煙酸(5.0 g,35.9 mmol)的無水乙醇(180 mL)溶液中緩慢加入濃硫酸(1.0 mL)。將反應(yīng)混合物于60℃下攪拌20分鐘,隨后加入額外的乙醇(33.0 mL),并將反應(yīng)體系加熱至回流狀態(tài),持續(xù)攪拌6.5小時(shí)。反應(yīng)完成后,將反應(yīng)液冷卻至0℃,并緩慢加入飽和碳酸氫鈉水溶液以中和反應(yīng)液。隨后,將乙醇通過旋轉(zhuǎn)蒸發(fā)儀減壓濃縮除去。用乙酸乙酯萃取水相,合并有機(jī)相,用飽和食鹽水洗滌,無水硫酸鈉干燥。過濾后,將濾液減壓濃縮,得到白色晶體狀產(chǎn)物6-羥基煙酸乙酯(5.33 g,收率89%)。產(chǎn)物經(jīng)1H-NMR(CDCl3)表征:δ 1.36(3H,t,J = 7.1 Hz),4.33(2H,q,J = 7.1 Hz),6.58(1H,d,J = 9.5 Hz),8.01(1H,dd,J = 2.4, 9.5 Hz),8.19(1H,d,J = 2.4 Hz),12.43(1H,brs)。
