a) 合成(4-氯苯基)-丙炔酸乙酯(Va): 在氬氣保護(hù)下,向裝有1-氯-4-碘苯(130.0 g,0.55 mol)、雙(三苯基膦)鈀(II)氯化物(7.57 g,10.8 mmol,2 mol%)、碘化亞銅(I)(4.19 g,22.0 mmol,4 mol%)的干燥四頸燒瓶中加入無水THF(1.4 L)。在室溫下,于5分鐘內(nèi)緩慢加入碳酸銫(355.3 g,1.09 mol,2當(dāng)量)。隨后,加入丙炔酸乙酯(111.3 mL,1.09 mol,2當(dāng)量),并將反應(yīng)混合物于35℃攪拌過夜。反應(yīng)完成后,補(bǔ)加丙炔酸乙酯(11.1 mL,0.11 mol,0.2當(dāng)量),繼續(xù)于35℃攪拌3小時(shí)。反應(yīng)混合物經(jīng)旋轉(zhuǎn)蒸發(fā)濃縮至干,殘余物溶于甲苯(0.5 L)和庚烷(1 L)中。所得懸浮液于40℃攪拌1小時(shí)后,通過硅藻土過濾。濾液濃縮后,經(jīng)硅膠柱層析(洗脫劑:甲苯/庚烷,1:2,v/v)純化,得到淺黃色固體產(chǎn)物Va(72.6 g,收率61%)。
