將10g 6-氧代庚酸(69.4mmol)溶解于100mL甲醇中。向溶液中加入幾滴濃硫酸作為催化劑,并在回流條件下攪拌反應(yīng)1.5小時(shí)。反應(yīng)完成后,通過(guò)旋轉(zhuǎn)蒸發(fā)儀將反應(yīng)混合物濃縮。將濃縮物溶解于二氯甲烷中,并用飽和碳酸氫鈉水溶液洗滌一次。分離有機(jī)相和水相,有機(jī)相用無(wú)水硫酸鈉干燥后,再次通過(guò)旋轉(zhuǎn)蒸發(fā)儀濃縮。最終獲得10.1g 5-乙酰戊酸甲酯,產(chǎn)率為理論值的91.1%。產(chǎn)物經(jīng)1H-NMR(400MHz,CDCl3)表征:δ=3.67(s,3H),2.44(t,2H),2.32(t,2H),2.13(s,3H),1.67-1.55(m,4H)。
