a)2-苯基-吡啶-4-基胺的合成:向16.0g(124mmol)2-氯-4-氨基吡啶溶于200ml甲苯的攪拌溶液中,依次加入18.2g(149mmol)苯基硼酸、7.19g(6.22mmol)四(三苯基膦)鈀(0)和130ml(260mmol)2M碳酸鈉水溶液。將反應(yīng)混合物于100℃下加熱回流16小時(shí),隨后冷卻至室溫。反應(yīng)液用乙酸乙酯萃取三次,合并有機(jī)相。有機(jī)相用200ml 1M鹽酸水溶液洗滌三次。合并酸水相,用5M氫氧化鈉水溶液調(diào)節(jié)至堿性,再用二氯甲烷萃取三次。合并有機(jī)萃取液,用無(wú)水硫酸鈉干燥,減壓濃縮。粗產(chǎn)物經(jīng)快速柱色譜純化(洗脫劑:丙酮),隨后在含少量乙醚的己烷中研磨,得到18.5g(收率87%)2-苯基-吡啶-4-基胺,為白色固體。ES-MS m/e(相對(duì)豐度):171([M+H]+,100)。
