以3-溴-5-氟苯甲酸為原料合成(3-溴-5-氟苯基)甲醇的一般步驟:將3-溴-5-氟苯甲酸(1.0 g,4.52 mmol)懸浮于四氫呋喃(8 mL)中,并將混合物冷卻至0℃。在15分鐘內(nèi)緩慢滴加硼烷四氫呋喃絡(luò)合物(1 M于四氫呋喃中,9 mL,9.0 mmol)。將反應(yīng)混合物逐漸升溫至室溫并攪拌過夜。反應(yīng)完成后,將混合物再次冷卻至0℃,并小心加入過量甲醇淬滅反應(yīng)。隨后,用乙酸乙酯稀釋反應(yīng)混合物,依次用1 N氫氧化鈉溶液(2×)和飽和鹽水(2×)洗滌。有機(jī)相用無水硫酸鈉干燥,過濾后減壓濃縮,得到0.88 g(產(chǎn)率95%)白色固體產(chǎn)物(3-溴-5-氟苯基)甲醇。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(CDCl3, 500 MHz)確認(rèn):δ 7.29(s, 1H),7.16(d, J = 7.9 Hz, 1H),7.03(d, J = 9.2 Hz, 1H),4.66(s, 2H)。
