將8-羥基喹啉(5g,0.034mol)溶解于乙醇(50mL)中,隨后加入溶于水(15mL)的氫氧化鈉(10g,0.25mol)。將混合溶液加熱回流,并在30分鐘內(nèi)緩慢滴加三氯甲烷。繼續(xù)回流反應(yīng)20小時(shí)后,通過減壓蒸餾去除乙醇和未反應(yīng)的三氯甲烷。將剩余物溶解于水(150mL)中,并用稀鹽酸調(diào)節(jié)pH至微酸性。所得黃色固體用二氯甲烷萃取,萃取液經(jīng)柱色譜(硅膠為固定相,二氯甲烷/甲醇為洗脫劑)純化,最終得到8-羥基喹啉-5-甲醛,為白色固體(115mg,0.67mmol)。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(CDCl3, 400MHz)和13C-NMR(DMSO-d6, 400MHz)確認(rèn):1H-NMR δ = 10.16(s, 1H), 9.71(dd, 1H, J = 1.6Hz, 8.4Hz), 8.89(dd, 1H, J = 1.6Hz, 4.0Hz), 8.02(d, 1H, J = 8.0Hz), 7.69(q, 1H, J = 4.0Hz), 7.30(d, 1H, J = 8.0Hz); 13C-NMR δ = 193.1, 160.5, 149.9, 141.1, 138.9, 133.9, 127.7, 125.5, 123.4, 111.7。
