步驟1:在0℃條件下,向含有3-甲氧基肉桂酸(11.0g,62mmol)和三乙胺(12.5g,124mmol)的丙酮(80mL)溶液中緩慢滴加氯甲酸乙酯(約1.5當(dāng)量)。維持該溫度反應(yīng)1小時(shí)后,滴加NaN3水溶液(6.40g,100mmol溶于35mL H2O中),并將反應(yīng)混合物于室溫下攪拌16小時(shí)。反應(yīng)完成后,加入水(100mL)并真空除去揮發(fā)性物質(zhì)。用甲苯(3×50mL)萃取反應(yīng)混合物,合并有機(jī)層,用MgSO4干燥后過(guò)濾。將濾液緩慢滴加至預(yù)熱的二苯基甲烷(50mL)和三丁胺(30mL)混合溶液中,滴加過(guò)程中通過(guò)蒸餾除去甲苯。滴加完畢后,將反應(yīng)溫度升至210℃并維持2小時(shí)。反應(yīng)結(jié)束后,冷卻混合物,過(guò)濾收集沉淀產(chǎn)物,用己烷(2×50mL)洗滌,干燥后得到6-甲氧基異喹啉-1(2H)-酮,為白色固體(5.53g,產(chǎn)率51%)(參考文獻(xiàn):Nicolas Briet等,Tetrahedron,2002,5761-5766)。LC-MS分析顯示MS m/z 176(M++H)。
