以2-苯基氨基亞甲基-丙二酸二乙酯為原料合成4-羥基喹啉-3-羧酸乙酯的一般步驟:在裝有機(jī)械攪拌器的1L三頸燒瓶中,加入2-苯基氨基亞甲基-丙二酸二乙酯(26.3g,0.100mol)、多磷酸(270g)和磷酰氯(750g)。將反應(yīng)混合物加熱至70℃,并在該溫度下攪拌4小時。反應(yīng)完成后,將混合物冷卻至室溫,然后進(jìn)行過濾。將濾得的殘余物用碳酸鈉(Na2CO3)水溶液處理,再次過濾,用水洗滌,最后干燥。得到4-羥基喹啉-3-羧酸乙酯,為淺棕色固體(15.2g,收率70%)。
