以2-氯-4-氨基嘧啶為原料合成4-氨基-2-甲氧基嘧啶的一般步驟如下: 預(yù)步驟A: 2-甲氧基-4-嘧啶胺的合成 將2-氯-4-嘧啶胺(150 mg,1.158 mmol)和甲醇鈉(188 mg,3.47 mmol)溶于甲醇(2.9 mL)中,將混合物在Biotage微波反應(yīng)器中加熱至100°C,反應(yīng)40分鐘。反應(yīng)完成后,通過反相高效液相色譜(RP-HPLC)純化粗產(chǎn)物,得到2-甲氧基-4-嘧啶胺(73 mg,0.583 mmol,收率50%)。質(zhì)譜(電噴霧正離子模式)m/z 126.0([M+H]+)。
