以3,7-二甲基-1-辛醇為原料合成(R)-1-溴-3,7-二甲基辛烷的一般步驟如下:在攪拌條件下,將濃硫酸(17 mL)緩慢加入48%的氫溴酸(100 mL)中,隨后加入3,7-二甲基-1-辛醇(67 mL,100 g)。將反應(yīng)混合物加熱至120-125℃,并在此溫度下維持3小時(shí)。反應(yīng)完成后,冷卻混合物,用庚烷(300 mL)進(jìn)行萃取。庚烷層依次用稀鹽酸、水和碳酸氫鈉溶液洗滌,經(jīng)干燥后濃縮,得到81.5 g油狀粗產(chǎn)物。將此粗產(chǎn)物在120-125℃的浴溫下進(jìn)行真空蒸餾,收集沸點(diǎn)為85-87℃/10 mmHg的餾分,得到無(wú)色油狀的目標(biāo)產(chǎn)物(R)-1-溴-3,7-二甲基辛烷,產(chǎn)量78.2 g,收率100%。質(zhì)譜分析顯示分子離子峰m/z 220,222(M+)。
