以4-氯-7-(3-氯丙氧基)-6-甲氧基喹啉-3-腈(31.2g,0.10mol)、2,4-二氯-5-甲氧基苯胺(21.1g,0.11mol)和吡啶鹽酸鹽(12.8g,0.11mol)為原料,在2-甲氧基乙醇(150g)中混合。將反應混合物加熱至120℃,并維持此溫度反應3小時。反應完成后,將懸浮液冷卻至室溫。在持續(xù)攪拌下,將反應混合物緩慢倒入水(600g)中。過濾收集生成的固體產(chǎn)物,并用水(2×40g)洗滌。隨后,將產(chǎn)物在50℃下干燥,得到7-(3-氯丙氧基)-4-[(2,4-二氯-5-甲氧基苯基)氨基]-6-甲氧基-3-氰基喹啉(35.9g,產(chǎn)率77%),為灰白色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR、13C NMR和質(zhì)譜(ESI)確認。1H NMR(400MHz,DMSO-d6)δ: 2.28(m,J = 6.0Hz,2H),3.82(t,J = 6.0Hz,2H),3.87(s,3H),3.97(s,3H),4.30(t,J = 6.0Hz,2H),7.35(s,1H),7.37(s,1H),7.75(s,1H),7.85(s,1H),8.43(s,1H),9.63(s,1H)。13C NMR(100MHz,DMSO-d6)δ: 31.9, 42.1, 56.7, 57.3, 65.8, 86.9, 102.4, 110.0, 113.1, 114.0, 117.4, 120.9, 123.6, 130.3, 136.7, 146.2, 149.8, 149.9, 151.3, 152.9, 154.5。質(zhì)譜(ESI)m/z: 468.0 [M + H]+。
