以2,5-二溴吡啶和二乙基鋅為原料合成2-乙基-5-溴吡啶的一般步驟:在0℃下,將2,5-二溴吡啶(10g,42.21mmol)和四(三苯基膦)鈀(Pd(PPh3)4,2.43g,2.11mmol)在四氫呋喃(THF,100mL)中的懸浮液中緩慢加入1M二乙基鋅(Et2Zn,42.2mL,42.21mmol)。反應(yīng)混合物在0℃下攪拌30分鐘,隨后升溫至室溫繼續(xù)攪拌1小時(shí)。反應(yīng)完成后,用飽和碳酸氫鈉(NaHCO3)溶液淬滅反應(yīng),并用乙酸乙酯(EtOAc)萃取。合并的有機(jī)層依次用水和飽和食鹽水洗滌,無水硫酸鈉(Na2SO4)干燥,過濾后減壓濃縮。粗產(chǎn)物通過硅膠柱色譜法純化,以4%乙酸乙酯/石油醚為洗脫劑,得到5-溴-2-乙基吡啶,為淺黃色液體(3g,收率38%)。薄層色譜(TLC)Rf值為0.6(10%乙酸乙酯/石油醚)。1H NMR(400MHz,CDCl3):δ 8.57(d,J=2.4Hz,1H),7.72-7.69(m,1H),7.06(d,J=8.0Hz,1H),2.81-2.75(m,2H),1.29(t,J=8.0Hz,3H)。
