4.2.7 順式-2-苯基-1,3-二惡烷-5-醇(18)的合成:在裝有Dean-Stark分水器的反應(yīng)瓶中,加入新蒸餾的苯甲醛(48 mL,50 g,0.47 mol)和甘油(44 mL,55 g,0.60 mol)的甲苯(69 mL)溶液,隨后滴加濃硫酸(3滴)。將反應(yīng)混合物加熱至回流,直至水分完全分離(收集8.5 mL水)。反應(yīng)完成后,將混合物冷卻至室溫,減壓蒸餾除去溶劑。得到的白色固體通過異丙醚/石油醚混合溶劑重結(jié)晶。收集沉淀物,并通過快速柱色譜法(色譜柱尺寸:?= 6 cm,h = 15 cm,洗脫劑體積V = 50 mL,環(huán)己烷/乙酸乙酯= 2:1,Rf = 0.16)進(jìn)一步純化,得到目標(biāo)產(chǎn)物18,為無色固體(42.6 g,0.24 mol,收率50%)。熔點(diǎn)(Mp)= 81℃;1H NMR(DMSO-d6)δ: 3.48-3.52(m,1H,OCH2CHOH),3.91-3.97(m,2H,OCH2CHOH),4.01-4.07(m,2H,OCH2CHOH),4.99(d,J = 4.8 Hz,1H,OH),5.53(s,1H,CHPh),7.33-7.40(m,3H,苯環(huán)3',4',5'-H),7.42-7.47(m,2H,苯環(huán)2',6'-H);13C NMR(DMSO-d6)δ: 62.4(OCH2CHOH),71.4(2C,OCH2CHOH),100.2(CHPh),126.2(2C,苯環(huán)C-2', C-6'),127.9(2C,苯環(huán)C-3', C-5'),128.5(苯環(huán)C-4'),138.9(苯環(huán)C-1');IR(純樣,cm-1): 3271, 2916, 2851, 1450, 1385, 1339, 1277, 1153, 1084, 995, 976, 806, 741, 694;HRMS(m/z): [M + H]+計(jì)算值C10H13O3,181.0859;實(shí)測(cè)值,181.0878;HPLC(方法1): tR = 11.7 min,純度95.4%。
