向裝有N,N-二甲基甲酰胺(DMF,100 mL)的三頸燒瓶中依次加入氰基乙酸乙酯(56.6 g,0.5 mol)、硫粉(16.0 g,0.5 mol)和三乙胺(50.5 g,0.5 mol),攪拌至完全溶解。隨后,緩慢滴加丙醛(31.9 g,0.55 mol)的乙醛(15 mL)溶液。反應過程中,內(nèi)部溫度自然升至60℃,并在該溫度下持續(xù)攪拌加熱。反應完成后,將反應混合物緩慢倒入水中,用乙酸乙酯進行萃取。合并有機相,依次用水和飽和食鹽水洗滌,無水硫酸鈉干燥后,減壓蒸餾除去溶劑。所得粗產(chǎn)物通過硅膠柱色譜法(MERCK Kiesel gel 60,洗脫劑為20%乙酸乙酯-正己烷)純化,得到2-氨基-5-甲基噻吩-3-羧酸乙酯(81.8 g,收率89%)。產(chǎn)物經(jīng)1H-NMR(400 MHz,CDCl3)表征:δ 1.33(t,3H,J = 7.2 Hz),2.26(d,3H,J = 1.2 Hz),4.25(q,2H,J = 7.2 Hz),5.76(br.s,2H),6.61(q,1H,J = 1.2 Hz)。
