2-甲基-2,3,4,5-四氫-1H-吡啶并[4,3-b]吲哚(17)的合成:將苯肼(1.0 g,9.3 mmol)和N-甲基-4-哌啶酮(1.1 g,9.3 mmol)溶解于1,4-二惡烷(35 mL)中,并將混合物冷卻至0℃。在0℃及攪拌條件下,緩慢滴加濃硫酸(5 mL),反應(yīng)過(guò)程中有沉淀生成。隨后,將反應(yīng)混合物加熱至60℃并維持1小時(shí),直至沉淀完全溶解。繼續(xù)在60℃下攪拌反應(yīng)混合物1小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,通過(guò)加入飽和碳酸氫鈉水溶液調(diào)節(jié)pH至約12,必要時(shí)加入少量固體氫氧化鈉輔助調(diào)節(jié)。用氯仿(3×20 mL)萃取有機(jī)產(chǎn)物,合并有機(jī)相后用鹽水(15 mL)洗滌,無(wú)水硫酸鈉干燥,減壓濃縮。通過(guò)柱色譜法純化(洗脫梯度:0-80%乙酸乙酯的己烷溶液),得到目標(biāo)產(chǎn)物(1.6 g,收率93%),為米色固體。1H NMR(400 MHz, DMSO-d6): δ 10.80(s, 1H), 7.30(m, 2H), 6.98(m, 2H), 3.53(s, 2H), 2.79(t, J = 5.2 Hz, 2H), 2.71(t, J = 5.4 Hz, 2H), 2.43(s, 3H)。13C NMR APT(100 MHz, CDCl3): δ 136.2(CH), 132.0(CH), 126.0(CH), 121.0(CH2), 119.1(CH2), 117.4(CH2), 110.7(CH2), 108.3(CH), 52.5(CH2), 51.8(CH2), 45.8(CH3), 23.5(CH2)。ESI-HRMS(m/z): [M + H]+ 計(jì)算值 C12H14N2, 187.1230; 實(shí)測(cè)值, 187.1228。
