a) 制備6-氯-N-甲基煙酰胺:將6-氯-3-吡啶甲酰氯(5.65 g,32.1 mmol)溶于四氫呋喃(64 mL)中,依次加入三乙胺(5.59 mL,40.1 mmol)和甲胺的四氫呋喃溶液(20.1 mL,40.1 mmol),反應(yīng)在冰浴條件下進(jìn)行。加畢,撤去冰浴,于室溫下攪拌反應(yīng)3.5小時(shí)。反應(yīng)完成后,將反應(yīng)液減壓濃縮,所得殘余物通過重結(jié)晶(乙酸乙酯/己烷)純化,得到6-氯-N-甲基-煙酰胺(5.06 g,收率92%)為淺棕色晶體。1H-NMR(CDCl3)δ:3.05(3H,d,J = 4.6 Hz),6.14(1H,brs),7.42(1H,d,J = 8.3 Hz),8.09(1H,dd,J = 2.4, 8.3 Hz),8.72(1H,d,J = 2.4 Hz)。
