以1-(5-溴-2-氟苯基)乙酮為原料合成5-溴-3-甲基-1H-吲唑的一般步驟:將1-(5-溴-2-氟苯基)乙酮(66.0 g,304 mmol)和350 mL無水肼加入1升圓底燒瓶中。將反應(yīng)混合物在117℃下回流5小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,減壓蒸發(fā)除去過量肼,得到白色固體。向固體中加入400 mL水,過濾分離水相。固體用400 mL水洗滌兩次。為去除痕量肼,將白色固體溶解于600 mL乙酸乙酯中,依次用300 mL水洗滌兩次和飽和鹽水洗滌。有機(jī)相用無水硫酸鈉干燥后,減壓濃縮溶劑,得到白色無定形固體產(chǎn)物5-溴-3-甲基-1H-吲唑(60.0 g,產(chǎn)率94%)。產(chǎn)物無需進(jìn)一步純化,可直接用于后續(xù)反應(yīng)。
