以對(duì)氨基苯乙酸(20g,0.132mol)和甘油(51g,0.555mol)為原料,合成6-喹啉乙酸的一般步驟如下:將氨基苯乙酸、FeSO4(4.9g,0.032mol)、甘油、PhNO2(10.5g,0.086mol)和濃H2SO4(23mL)混合,加熱至200℃并維持5小時(shí)。反應(yīng)完成后,用4N NaOH水溶液調(diào)節(jié)反應(yīng)混合物的pH至10。過(guò)濾反應(yīng)混合物,將澄清的濾液用AcOH酸化至pH約5。過(guò)濾收集生成的沉淀,得到6-喹啉乙酸(12.5g,產(chǎn)率51%),為棕色固體。TLC條件:20% MeOH/CH2Cl2,Rf=0.2。1H NMR(300MHz,DMSO-d6)δ ppm:8.85(m,1H),8.31-8.28(m,1H),7.96(d,J=8.3Hz,1H),7.82(m,1H),7.67-7.64(m,1H),7.50-7.49(m,1H),3.78(s,2H)。
