以2-硝基-3-氯苯酚為原料合成2-氨基-3-氯苯酚的一般步驟:在室溫條件下,將3-氯-2-硝基苯酚(2.5g,14.5mmol)溶于乙醇(30mL)中,隨后加入二水合氯化亞錫(SnCl2·2H2O,13g,57.8mmol)。將反應(yīng)混合物加熱至90°C并持續(xù)攪拌2小時。通過薄層色譜(TLC)監(jiān)測反應(yīng)進(jìn)度,確認(rèn)反應(yīng)完成后,將反應(yīng)混合物冷卻至室溫,并在減壓條件下濃縮。向濃縮后的殘余物中加入冰水(50mL),并用氨水溶液調(diào)節(jié)pH至7。隨后,用乙酸乙酯(3×50mL)進(jìn)行萃取。合并有機層,用飽和食鹽水(50mL)洗滌,無水硫酸鈉(Na2SO4)干燥,過濾后減壓濃縮。將得到的殘余物用正己烷(25mL)研磨,得到灰白色固體2-氨基-3-氯苯酚。產(chǎn)量:1.8g(收率80%)。產(chǎn)物經(jīng)1H NMR(400MHz,DMSO-d6)和質(zhì)譜(ESI+)表征:1H NMR δ 9.58(寬單峰,1H),6.69(雙峰,J = 8.0Hz,1H),6.63(雙峰,J = 8.0Hz,1H),6.38-6.48(多重峰,1H),4.05(寬單峰,2H);質(zhì)譜(ESI+)m/z 144.09 [M + H]+。
