步驟A:1-甲基-1H-1,2,4-三唑-3-甲酸甲酯的制備:在氮氣保護下,向0℃攪拌中的氫化鈉(60%油分散體,0.346g,8.66mmol)在N,N-二甲基甲酰胺(DMF,20mL)中的懸浮液中,緩慢滴加1H-1,2,4-三唑-3-羧酸甲酯(1.00g,7.87mmol)的DMF(20mL)溶液。反應(yīng)混合物在0℃下繼續(xù)攪拌1小時。隨后,逐滴加入碘甲烷(MeI,0.982mL,15.7mmol)。將反應(yīng)混合物在室溫下攪拌過夜。反應(yīng)完成后,將混合物倒入冷水中,并用乙酸乙酯(EtOAc)萃取。合并有機層,用飽和食鹽水洗滌,無水硫酸鈉干燥,減壓濃縮。殘余物通過硅膠柱色譜(洗脫劑:己烷/乙酸乙酯=3:1)純化,得到目標(biāo)產(chǎn)物1-甲基-1H-1,2,4-三唑-3-甲酸甲酯(0.380g,收率34%),為白色固體。質(zhì)譜(APCI)m/z = 142.1(M + H)。
