以三甲硅基丙炔醇為原料合成反-3-(三甲基硅基)烯丙醇的一般步驟:在氬氣保護(hù)下,將實(shí)施例40中制備的醇(1.54 g,12 mmol,1當(dāng)量)溶于無(wú)水THF(12 mL)中,緩慢滴加到LiAlH4(0.7 g,18 mmol,1.5當(dāng)量)在THF(15 mL)中的懸浮液中,室溫?cái)嚢?。隨后,將反應(yīng)混合物加熱回流4小時(shí)。反應(yīng)完成后,用飽和NaHCO3水溶液和NH4Cl水溶液淬滅反應(yīng)。用Et2O(3 × 50 mL)萃取反應(yīng)混合物,合并有機(jī)層,用鹽水洗滌,經(jīng)MgSO4干燥。減壓濃縮除去溶劑,得到目標(biāo)產(chǎn)物(1.39 g,10.6 mmol,收率89%)為無(wú)色液體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400 MHz, CDCl3)δ 6.19(1H, m, H-3),5.93(1H, dt, J2-3 = 18.8 Hz, J2-4 = 1.45 Hz, H-2),4.18(2H, dd, J4-3 = 2.72, J4-2 = 1.45, H-4),1.56(1H, s, H-5),0.09(9H, s, H-1);13C NMR(100 MHz, CDCl3)δ 144.8(C-3),129.5(C-2),65.5(C-4),-1.37(C-5);IR(純, cm-1)3331(b, OH),1622(s, C=C)確認(rèn)。
