以N,N-二甲基-2-(2-甲基-4-吡啶基)乙胺(6.4 g,39 mmol)為原料,將其甲醇溶液(25 mL)緩慢滴加至高碘酸鈉(25.2 g,117 mmol)的甲醇(25 mL)懸浮液中,反應(yīng)在室溫下進(jìn)行。滴加完畢后,繼續(xù)在室溫下攪拌反應(yīng)混合物1小時(shí)。反應(yīng)完成后,過(guò)濾去除不溶性沉淀,濃縮濾液。將濃縮后的殘余物與水混合,用乙酸乙酯進(jìn)行萃取。合并有機(jī)層,用飽和鹽水洗滌,無(wú)水硫酸鎂干燥。減壓蒸發(fā)溶劑,得到目標(biāo)產(chǎn)物2-甲基-4-吡啶甲醛(3.7 g,收率78%)。產(chǎn)物經(jīng)1H-NMR(CDCl3)表征:δ 2.68(3H,s),7.51(1H,d,J = 4.9 Hz),7.56(1H,s),8.76(1H,d,J = 4.9 Hz),10.05(1H,s)。
