以2-[(4-甲氧基苯胺基)亞甲基]丙二酸二乙酯(CAS: 83507-70-4,71.4 g,0.24 mol)為原料,在150 mL二苯醚中制備溶液。將反應(yīng)體系置于金屬浴中,緩慢加熱至243℃,并維持此溫度反應(yīng)3小時。反應(yīng)過程中,蒸餾出15 mL液體。反應(yīng)完成后,將混合物冷卻至室溫,隨后加入250 mL己烷溶解。攪拌混合物3小時后,進(jìn)行過濾操作。用己烷洗滌所得固體,干燥后得到4-羥基-6-甲氧基喹啉-3-羧酸乙酯(25.0 g,0.10 mol,產(chǎn)率41%),其純度滿足直接用于后續(xù)反應(yīng)的要求。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(CDCl3)和LC-MS確認(rèn):1H-NMR(CDCl3)δ= 1.28(t,3H),3.85(s,3H),4.21(q,2H),7.34(dd,1H),7.55-7.62(m,2H),8.49(s,1H);LC-MS:Rt = 1.17 min;MS:m/z = 248 [M + 1]+。
