以6-氧代-1-苯基-1,6-二氫-吡啶-3-羧酸甲酯為原料合成6-氧代-1-苯基-1,6-二氫吡啶-3-甲酸的一般步驟:在0℃下,將氫氧化鋰一水合物(0.366 g,8.73 mmol)加入到含有6-氧代-1-苯基-1,6-二氫吡啶-3-羧酸甲酯(1.0 g,4.37 mmol)的四氫呋喃(9 mL)和水(6 mL)混合溶液中,攪拌反應(yīng)1小時(shí)。反應(yīng)完成后,用水稀釋反應(yīng)混合物,并用乙酸乙酯洗滌。隨后,用2N鹽酸將水層的pH調(diào)節(jié)至2,過濾收集沉淀物,得到6-氧代-1-苯基-1,6-二氫吡啶-3-甲酸,為棕色固體(0.740 g,收率79%)。產(chǎn)物熔點(diǎn)為256-263°C。產(chǎn)物結(jié)構(gòu)通過1H NMR(400 MHz,DMSO-d6)δ 6.53(d,J = 9.4 Hz,1H),7.40-7.49(m,5H),7.87(dd,J = 2.5, 9.8 Hz,1H),8.23(d,J = 2.5 Hz,1H);IR(KBr)ν 3446, 1708, 1645, 1577, 1263, 1228 cm-1;MS 214(M-1)進(jìn)行表征。
