在氬氣保護(hù)下,向攪拌中的2-氯-3-氧代丙酸乙酯(5.0 g,33 mmol)的苯(50 mL)溶液中加入硫代乙酰胺(2.5 g,33 mmol),并將混合物加熱回流。反應(yīng)4小時后,將反應(yīng)混合物冷卻至室溫,用水(50 mL)稀釋,并用飽和碳酸氫鈉溶液中和至pH 7。隨后,用乙酸乙酯萃取反應(yīng)混合物,合并有機(jī)相,依次用水和鹽水洗滌。有機(jī)層經(jīng)無水硫酸鈉干燥后,過濾并真空濃縮。通過硅膠柱色譜純化(洗脫劑:庚烷/乙酸乙酯,0至50%梯度),得到2-甲基噻唑-5-羧酸乙酯(2.68 g,47%收率),為黃色液體。質(zhì)譜(MS)分析顯示:m/e = 172.0 [M + H]+。
