在3頸5L圓底燒瓶中,配備機(jī)械攪拌器、J-KEM溫度探針及連接至鼓泡器的N2入口適配器。向燒瓶中加入甘氨酸芐酯鹽酸鹽(505.2g,2.51mol,1.0當(dāng)量)和二氯甲烷(3.0L)。隨后加入二苯甲酮亞胺(471.1g,97%純度,2.6mol,1.00當(dāng)量)至乳白色反應(yīng)混合物中,觀察到反應(yīng)放熱(溫度上升4.5℃)。將反應(yīng)混合物于20℃下攪拌3小時,通過TLC(展開劑:50%乙酸乙酯/庚烷)檢測顯示僅有痕量起始原料殘留。補加二苯甲酮亞胺(25.0g,0.14mol)后,繼續(xù)在20℃下攪拌15小時,TLC確認(rèn)反應(yīng)完成。反應(yīng)混合物經(jīng)短硅藻土墊過濾以除去生成的氯化銨,濾餅用二氯甲烷(1.5L)洗滌。合并濾液后,減壓濃縮得到白色固體,進(jìn)一步真空干燥獲得粗產(chǎn)物N-二苯亞甲基甘氨酸芐酯878.7g(產(chǎn)率106%)。產(chǎn)物經(jīng)1H-NMR(DMSO-d6)鑒定:δ 7.53-7.25(m, 13H),7.12(dd, 2H),5.10(s, 2H)和4.17(s, 2H)。HPLC分析顯示純度>95%。
