向攪拌的3-氧雜-9-氮雜雙環(huán)[3.3.1]壬烷-7-酮鹽酸鹽(1.26 g, 7.08 mmol, CAS: 1126795-00-3, 購自PharmaBlock (Nanjing) R&D Co., Ltd.)的DMF(10 mL)溶液中,室溫下緩慢滴加碳酸鉀(2.94 g, 14.16 mmol)的DMF(10 mL)溶液。隨后加入溴化芐(1.33 g, 7.78 mmol)。將反應混合物在室溫下攪拌過夜。反應完成后,將混合物濃縮,殘余物用水(5 mL)和乙酸乙酯(20 mL)分配。分離有機層,用無水硫酸鈉干燥,過濾后濃縮。通過柱層析純化殘余物,得到目標化合物9-芐基-3-氧雜-9-氮雜雙環(huán)[3.3.1]壬-7-酮(1.38 g, 84.2%收率)。產(chǎn)物結(jié)構經(jīng)1H NMR(CDCl3, 400 MHz)和LC/MS確認:1H NMR (CDCl3, 400 MHz) δ 7.36 (m, 5H), 3.93 (s, 2H), 3.86 (d, J = 11.0 Hz, 2H), 3.73 (d, J = 10.8 Hz, 2H), 3.18 (d, J = 5.6 Hz, 2H), 2.76 (dd, J = 15.9, 5.6 Hz, 2H), 2.35 (d, J = 15.6 Hz, 2H); LC/MS: 計算值232 (MH+), 實驗值232 (MH+)。
