以3-碘-1H-吡唑并[3,4-d]嘧啶-4-胺(5g,19.2mmol,1.0當(dāng)量)和2-溴丙烷(1.9mL,20.1mmol,1.05當(dāng)量)為原料合成3-碘-1-異丙基-1H-吡唑[3,4-d]并嘧啶-4-胺的一般步驟如下:將3-碘-1H-吡唑并[3,4-d]嘧啶-4-胺置于三頸燒瓶中,加入40mL DMF和碳酸鉀(5.3g,38.4mmol,2.0當(dāng)量)?;旌衔镉玫?dú)庵脫Q3次后,加入2-溴丙烷。將反應(yīng)混合物加熱至80℃,反應(yīng)4小時(shí)后通過TLC監(jiān)測(cè)反應(yīng)進(jìn)程。反應(yīng)完成后,減壓蒸餾除去DMF。殘余物用二氯甲烷(DCM)和水的混合溶劑萃取3次,合并DCM層并蒸發(fā)至干。產(chǎn)物通過從乙酸乙酯(EA)和石油醚(PE)(體積比1:3)的混合溶劑中重結(jié)晶純化,得到淡黃色粉末狀目標(biāo)化合物。重結(jié)晶母液中的產(chǎn)物通過柱色譜法進(jìn)一步純化,得到5.4g產(chǎn)物,收率92.8%。產(chǎn)物經(jīng)1H NMR(400MHz,DMSO-d6)表征:δ8.19(s,1H),4.99-4.93(m,1H),1.42(d,J=6.7Hz,6H)。質(zhì)譜(ESI)顯示m/z:304.0 [M+H]。
