以1-三苯甲基-4-溴吡唑和聯(lián)硼酸頻那醇酯為原料,合成4-(4,4,5,5-四甲基-1,3,2-二氧雜硼烷-2-基)-1-三苯甲基-1H-吡唑的一般步驟如下:在攪拌條件下,將5.0g(12.8mmol)1-三苯甲基-4-溴-1H-吡唑、9.8g(38.6mmol,3.0當(dāng)量)雙(頻哪醇合)二硼、1.0g(1.3mmol,10mol%)PdCl2(dppf)和6.3g(64.2mmol,5.0當(dāng)量)乙酸鉀溶于30mL DMF中,于80℃加熱反應(yīng)16小時(shí)。反應(yīng)完成后,將混合物用水稀釋,用二氯甲烷萃取產(chǎn)物。有機(jī)層依次用鹽水洗滌,無水硫酸鈉干燥,最后通過快速色譜法純化,得到5.1g(產(chǎn)率91%)目標(biāo)化合物4-(4,4,5,5-四甲基-[1,3,2]二氧雜硼雜環(huán)戊烷-2-基)-1-三苯甲基-1H-吡唑。
