向3-芐氧基-5-溴吡啶N-氧化物(9g,32.13mmol)的無水二氯甲烷溶液中緩慢滴加三氯氧磷(30mL,327.72mmol),將反應混合物加熱回流2小時。反應完成后,在減壓下蒸除溶劑,將殘余物小心倒入飽和碳酸鈉水溶液中。過濾收集析出的固體,用冷水洗滌,干燥后得到5-溴-2-3-芐氧基吡啶(9.10g,收率95%),為米色固體。熔點:76-77℃;IR(KBr,cm-1):1547, 1415, 1266, 1071;1H NMR(400MHz, CDCl3):δ 5.34(s, 2H, -OCH2Ph),7.41-7.52(m, 5H, Ph-H),8.03(d, J = 2.0Hz, 1H, 吡啶-H),8.19(d, J = 2.0Hz, 1H, 吡啶-H);13C NMR(100MHz, CDCl3):δ 70.8, 119.2, 124.6, 127.7, 128.3, 128.6, 135.5, 138.4, 140.8, 150.7;MS(ESI)m/z(相對豐度):297.8 [(M + H)+, 72%], 299.8 [(M + H + 2)+, 100%], 301.8 [(M + H + 4)+, 27%]。元素分析(C12H9BrClNO)計算值:C, 48.27; H, 3.04; N, 4.69。實測值:C, 48.37; H, 3.05; N, 4.71。
