以3-硝基-1H-吡唑?yàn)樵虾铣?-溴-3-硝基吡唑的一般步驟:在室溫條件下,將水(15 mL)加入至含有3-硝基-1H-吡唑(48a)(1 g,8.90 mmol)和N-溴代琥珀酰亞胺(NBS)(1.73 g,9.70 mmol)的混合體系中,持續(xù)攪拌反應(yīng)混合物過(guò)夜。通過(guò)薄層色譜(TLC)監(jiān)測(cè)反應(yīng)進(jìn)程,確認(rèn)反應(yīng)完全后,將反應(yīng)液用水稀釋?zhuān)S后用乙酸乙酯進(jìn)行萃取。有機(jī)相依次用飽和氯化鈉溶液洗滌,無(wú)水硫酸鈉干燥,減壓濃縮,經(jīng)旋轉(zhuǎn)蒸發(fā)除去溶劑,得到目標(biāo)化合物4-溴-3-硝基吡唑(48b)(1.66 g,產(chǎn)率93%,白色固體)。質(zhì)譜(ESI)數(shù)據(jù):m/z 192 [M + H]+。
