以2-氰基-6-甲氧基苯并噻唑?yàn)樵虾铣?-氰基-6-羥基苯并噻唑的一般步驟:在氬氣保護(hù)下,將吡啶鹽酸鹽(2.32 g)加入至2-氰基-6-甲氧基苯并噻唑(51.4 mg,0.271 mmol)中,加熱至200℃使吡啶鹽酸鹽完全溶解,持續(xù)攪拌反應(yīng)混合物30分鐘。反應(yīng)完成后,冷卻反應(yīng)混合物,隨后加入1M鹽酸溶液(50 mL)。用乙酸乙酯(3×50 mL)進(jìn)行萃取,合并有機(jī)層并用無(wú)水硫酸鈉干燥。干燥后的有機(jī)層經(jīng)減壓濃縮,得到粗產(chǎn)物。通過制備型薄層硅膠色譜法純化(使用20 cm×20 cm×1.75 mm的硅膠板,展開劑為己烷-乙酸乙酯(1:1)),得到2-氰基-6-羥基苯并噻唑(47.2 mg,收率99%)為淺黃色固體。1H NMR(270 MHz,CD3OD)δ 7.17(1H,dd,J = 2.7, 9.2 Hz),7.41(1H,d,J = 2.7 Hz),7.99(1H,d,J = 9.2 Hz)。
