Background

Retigabine (CAS 150812-12-7) is a positive allosteric modulator of neuronal potassium channels KCNQ2-5 (K_V7.2-7.5). By enhancing channel opening, retigabine stabilizes neuronal membrane potentials, thereby reducing excitability. Reported EC_50 values are 1.4 μM for KCNQ3/5, 1.6 μM for KCNQ2/3, and 5.2 μM for KCNQ4 subunits. Due to this activity, retigabine serves as an anticonvulsant agent in epilepsy research. Additionally, mutations in KCNQ2/3 channels are linked clinically to benign familial neonatal convulsions, highlighting retigabine's relevance in investigating neuronal channelopathies and seizure mechanisms.

0.1, 0.3, 1 or 3?μM retigabine for 4 h incubation

Applications

Retigabine reduced the KV2.1 current density in a concentration-dependent manner. The current density was significantly reduced by approximately 2.5-fold after exposure to 1 and 3 μM retigabine. Thus, KV2.1 was identified as a new molecular target for retigabine, with the potential to further explain retigabine's neuroprotective properties.

Applications

Retigabine exhibited an additive effect when administered in combination with classical anticonvulsants, most notably diazepam, phenobarbital, phenytoin as well as valproate.
