名稱	Favipiravir
描述	Favipiravir is a potent and selective RNA-dependent RNA polymerase inhibitor for the treatment of influenza virus infections.
細(xì)胞實(shí)驗(yàn)	The cytotoxicity of T-705 is evaluated by an assay with XTT. XTT is converted to aqueous formazan by an enzyme in MDCK cells, Vero cells, HEL cells, A549 cells, HeLa cells, and HEp-2 cells. The compounds are diluted to the appropriate concentrations (volume, 100 μl) with test medium (EMEM containing 10% FCS) in 96-well culture plates in which each well contains a concentration of 2 × 103 cells/100 μL. The test plates are incubated for 3 days at 37°C in 100% humidity and 5% CO2. After 3 days, 50 μl of the XTT reagent (1 mg/ml in FCS-free EMEM containing 5 mM phenazine methosulfate) is added, and the reaction product is assayed by measurement of the absorbance at 450 nm with a microplate reader. Cytotoxicity is expressed as the 50% cell-inhibitory concentration (CC50).(Only for Reference)

體內(nèi)活性	方法：當(dāng)Favipiravir (T-705)使用劑量為1-100mg/kg，并在感染致死劑量的流感病毒 A/Victoria/3/75（H3N2）、A/Osaka/5/70（H3N2）或 A/Duck/MN/1525/81（H5N1）的小鼠中每天口服 2 或 4 次，持續(xù) 5 天時，觀察小鼠的生存率。結(jié)果：與安慰劑相比，在Favipiravir (T-705)在30 mg/kg/天或更高的劑量下顯示出更高的生存率,該藥物還以33mg/kg/天或更高的劑量對A/Duck/MN/1525/81（H5N1）病毒提供了顯著的保護(hù)。[1] 方法：在I型干擾素受體敲除（IFNAR ?/?）小鼠中，在致命的 SFTSV 感染后的第1-5天內(nèi)，以120mg / kg /天或200mg / kg /天的速度給予Favipiravir (T-705)，并持續(xù)5天，觀察小鼠的生長狀態(tài)。結(jié)果：當(dāng)在感染后 3 天和 4 天內(nèi)開始治療時，所有劑量為 120 mg/kg/天或 200 mg/kg/天的法匹拉韋治療的小鼠分別從致命的 SFTSV 感染中存活下來。[3]
