General procedure for the synthesis of 6-(trifluoromethyl)pyridine-3-carboxaldehyde from 6-(trifluoromethyl)-3-pyridine methanol:
Intermediate 174: 6-(Trifluoromethyl)nicotine derivative; 800 mg of 6-(trifluoromethyl)pyridin-3-yl)methanol (Intermediate 173, 4.52 mmol) was dissolved in 50 mL of dichloromethane (DCM) and cooled to 0°C. To this solution was added 2 g of manganese dioxide (MnO2, 23 mmol). The reaction mixture was stirred at room temperature overnight. Upon completion of the reaction, the insoluble material was removed by filtration and the filtrate was concentrated at room temperature to give 750 mg (95% yield) of 6-trifluoromethylpyridine-3-carbaldehyde as a white solid. The product was characterized by 1H-NMR (CDCl3): δ 7.88-7.90 (1H, d), 8.36-8.38 (1H, d), 9.20 (1H, s), 10.22 (1H, s); TLC (DCM/MeOH 10:1) showed Rf = 0.9. | [References]
