以2-氨基-6-氯-4-羥基嘧啶為原料合成2-氨基-4-氯-5-硝基-6-羥基嘧啶的一般步驟:在40℃條件下,將2-氨基-6-氯嘧啶-4(3H)-酮(10g,68.73mmol)溶于98%硫酸(46mL)中,保持?jǐn)嚢琛kS后,緩慢滴加65%硝酸(9g)至上述溶液中。反應(yīng)混合物在室溫下繼續(xù)攪拌1小時(shí)。反應(yīng)完成后,將溶液小心倒入100g冰中淬滅反應(yīng)。通過過濾收集沉淀,所得濾餅在減壓烘箱中干燥,得到12g(產(chǎn)率87%)目標(biāo)產(chǎn)物2-氨基-6-氯-5-硝基嘧啶-4(3H)-酮,為黃色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(300MHz,DMSO-d6)確認(rèn):δ7.13(1H,s),8.59(1H,s),12.16(1H,s)。
