一般步驟：向冷卻至0-5℃并攪拌的2.76 g（12.5 mmol）6-氨基青霉烷酸（6-APA）溶于60 mL水（含5.25 g，62.5 mmol碳酸氫鈉）的溶液中，緩慢滴加2.76 g（16.2 mmol）苯氧乙酰氯溶于5 mL丙酮的溶液，滴加時(shí)間控制在1分鐘內(nèi)。反應(yīng)混合物在10-15℃下劇烈攪拌20分鐘。反應(yīng)完成后，用15 mL甲基異丁基酮（MIBK）萃取水相兩次，棄去有機(jī)相。將水相冷卻至5-10℃，用冷的5.0 mol/L硫酸溶液酸化至pH 2.0。酸化后的水相用50 mL MIBK萃取兩次，合并有機(jī)相，用冷水洗滌，無水硫酸鈉干燥10分鐘。過濾除去干燥劑，向?yàn)V液中加入10 mL 25% 2-乙基己酸鉀的丁醇溶液，析出白色結(jié)晶。過濾收集固體，用干燥丙酮洗滌，真空干燥，得到3.5 g（產(chǎn)率80%）目標(biāo)化合物(2S,5R,6R)-3,3-二甲基-7-氧代-6-(2-苯氧基乙酰胺基)-4-硫雜-1-氮雜二環(huán)[3.2.0]庚-2-羧酸鉀，為白色固體。熔點(diǎn)：210-211℃（分解）。1H NMR (400 MHz, DMSO-d6): δ 8.42 (d, 1H, J = 8.0 Hz), 7.27 (m, 2H), 6.92 (m, 3H), 5.42 (dd, 1H, J = 8.0, 4.0 Hz), 5.40 (d, 1H, J = 4.0 Hz), 4.62 (d, 2H, J = 2.2 Hz), 3.88 (s, 1H), 1.52 (s, 3H), 1.46 (s, 3H); 13C NMR (100 MHz, DMSO-d6): δ 172.8, 169.0, 168.1, 158.1, 130.0, 121.7, 115.0, 74.6, 67.3, 66.7, 65.0, 57.8, 32.7, 27.8; HRMS (FAB) 計(jì)算值 C16H18KN2O5S [M + H]+: m/z 389.0584; 實(shí)測值: 389.0995。
