步驟1:以6-氯吡啶-3-醇(112g,868mmol)為原料,將其溶解于THF(800mL)和水(800mL)的混合溶劑中。隨后加入碳酸鈉(92.0g,1.736mol)和碘(244.2g,1.04mol)。將反應(yīng)混合物在室溫下攪拌4小時。反應(yīng)完成后,分離水相和有機(jī)相,產(chǎn)物主要存在于水相中。用水相產(chǎn)物經(jīng)己烷(400mL×2)洗滌以去除雜質(zhì)。接著,用鹽酸將水相的pH值調(diào)節(jié)至7,然后用乙酸乙酯(500mL×4)進(jìn)行萃取。合并有機(jī)層,用無水硫酸鈉干燥,過濾。濾液經(jīng)減壓濃縮得到粗產(chǎn)物,粗產(chǎn)物用乙酸乙酯洗滌,最終得到6-氯-2-碘吡啶-3-醇163g,產(chǎn)率為75%。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(DMSO,400MHz)確認(rèn):δ11.13(s,1H,OH),7.31(dd,J = 8.4,12.8Hz,2H)。
