在-78℃下,將正丁基鋰(n-BuLi,2.80 mL,2.60 M,7.27 mmol)的己烷溶液緩慢滴加到3-丁炔-1-醇(0.50 mL,6.61 mmol)的四氫呋喃(THF,7.5 mL)溶液中。反應(yīng)混合物在-78℃下保持5分鐘后,升溫至0℃并繼續(xù)攪拌30分鐘。隨后,加入對甲苯磺酰氯(p-TsCl,1.51 g,7.93 mmol)的THF(8 mL)溶液,反應(yīng)在0℃下繼續(xù)攪拌30分鐘。反應(yīng)完成后,加入水(2 mL)淬滅反應(yīng),混合物用乙醚(Et2O,3×10 mL)萃取。合并有機相,用無水硫酸鈉(Na2SO4)干燥,減壓濃縮除去溶劑,得到對甲苯磺酸3-丁炔酯(1.41 g,產(chǎn)率95%)為淺黃色油狀物。產(chǎn)物經(jīng)核磁共振氫譜(1H NMR,400 MHz,CDCl3)確認(rèn):δ 7.78(d,J = 8.0 Hz,2H),7.33(d,J = 8.0 Hz,2H),4.08(t,J = 7.1 Hz,2H),2.53(dt,J = 7.1, 2.6 Hz,2H),2.43(s,3H),1.95(t,J = 2.6 Hz,1H);核磁共振碳譜(13C NMR,100 MHz,CDCl3):δ 145.0, 132.7, 129.9, 127.9, 78.3, 70.7, 67.4, 21.6, 19.4;紅外光譜(IR,純):3290, 2962, 2919, 1598, 1359, 1190, 1176, 980, 904, 815 cm-1;質(zhì)譜(ESI):m/z 247.0399 [C11H12O3S(M+23)理論值247.0400],471(堿),247, 225。
