A) 在氮氣保護下,將甲醇(5.4 mL)緩慢滴加至含有60%氫化鈉(8.05 g)的四氫呋喃(THF,200 mL)懸浮液中,保持反應(yīng)溫度為0℃。隨后,在0℃下向反應(yīng)體系中加入4,6-二氯嘧啶(20.0 g)的THF(45 mL)溶液,繼續(xù)在0℃下攪拌反應(yīng)1小時。反應(yīng)完成后,在0℃下向反應(yīng)混合物中緩慢加入1N鹽酸進行中和,隨后用乙酸乙酯萃取反應(yīng)產(chǎn)物。有機相依次用水和飽和食鹽水洗滌,并用無水硫酸鎂干燥。減壓濃縮去除溶劑,殘余物通過己烷重結(jié)晶,得到目標(biāo)產(chǎn)物4-氯-6-甲氧基嘧啶(10.2 g),為淺黃色晶體。產(chǎn)物經(jīng)1H NMR(300 MHz,DMSO-d6)表征:δ 3.96(3H,s),7.19(1H,s),8.69(1H,s)。
