步驟A:在干燥的圓底燒瓶中依次加入Boc-L-纈氨酸(1.03 g,4.74 mmol)、N-羥基琥珀酰亞胺(1.22 g,10.6 mmol)和1-乙基-3-(3-二甲氨基丙基)碳二亞胺鹽酸鹽(EDC,1.60 g,8.35 mmol)。將上述試劑溶于無水二氯甲烷(30 mL)中,反應(yīng)體系通過橡膠隔膜密封,并用氬氣置換三次以排除氧氣。隨后,將反應(yīng)混合物在室溫下攪拌反應(yīng)。3天后,薄層色譜(TLC)檢測(cè)(茚三酮顯色)確認(rèn)Boc-L-纈氨酸已完全反應(yīng)。反應(yīng)液依次用水、飽和碳酸氫鈉水溶液洗滌,水相用二氯甲烷反萃取。合并有機(jī)相,用飽和食鹽水洗滌,無水硫酸鈉干燥,過濾。濾液經(jīng)減壓濃縮后,真空干燥得到白色固體Boc-L-纈氨酸羥基琥珀酰亞胺酯(1.52 g,產(chǎn)率100%)。產(chǎn)物經(jīng)1H-NMR(300 MHz,CDCl3)表征:δ 4.98(寬峰,1H),4.58(雙峰,1H),2.82(多重峰,4H),2.27(多重峰,1H),1.44(單峰,9H),1.03(雙峰,6H)。
