以3-硝基苯甲酰胺為原料合成3-氨基苯甲酰胺的一般步驟如下:將3-硝基苯甲酰胺(33.3g,198.8mmol)和FeCl3·6H2O(13.6g,50.4mmol)溶解于乙醇(600mL)中,加入活性炭(15g)。將反應(yīng)混合物在油浴中加熱至60-65℃。在40分鐘內(nèi),通過滴液漏斗緩慢加入肼(30mL,85%,510mmol)。加畢,繼續(xù)在60-65℃下攪拌反應(yīng)5小時。反應(yīng)完成后,減壓過濾混合物,濃縮濾液。向濃縮液中加入二氯甲烷(400mL),析出沉淀,過濾除去沉淀,濃縮濾液得到白色固體。該固體通過從乙酸乙酯(EA)中重結(jié)晶進(jìn)一步純化,最終得到3-氨基苯甲酰胺(21.0g,收率76%)為白色固體。產(chǎn)物經(jīng)1H NMR(DMSO-d6, 400MHz)和質(zhì)譜(ESI+)表征:1H NMR δ 8.10(s, 1H), 7.85-7.77(m, 1H), 7.76(d, J = 1.8Hz, 1H), 7.59-7.37(m, 3H), 3.63(s, 3H);MS(ESI+)m/z 137.1 [M + H]+。
