在氬氣保護下,將溴氧化磷(4 g,14.2 mmol)加熱至90℃,直至固體完全溶解。向該熱溶液中加入2,8-雙(三氟甲基)-4-羥基喹啉(4.08 g,14.2 mmol),并將油浴溫度升至150℃。反應6小時后,將混合物冷卻至室溫。通過加入冰水淬滅反應,過濾生成的沉淀,并用冷水洗滌,得到目標產(chǎn)物4-溴-2,8-二(三氟甲基)喹啉(4.70 g,收率96%),為白色固體。薄層色譜(TLC)Rf值為0.79(展開劑:環(huán)己烷/乙醚=5:1)。熔點:60℃。1H NMR(300 MHz,CDCl3)δ 7.82(t,J=7.9 Hz,1H),8.11(s,1H),8.22(d,J=7.3 Hz,1H),8.46(d,J=8.6 Hz,1H)。13C NMR(125 MHz,CDCl3)δ 120.9(q,J=276.0 Hz),122.0(q,J=2.0 Hz),123.6(q,J=273.8 Hz),128.9,129.4,129.8(q,J=30.8 Hz),130.5(q,J=5.3 Hz),131.5,138.5,144.5,148.6(q,J=36.1 Hz)。紅外光譜(IR)νmax:1577,1422,1302,1136,1098,1010,876,824 cm-1。氣相色譜-質譜聯(lián)用(GC-MS)m/z:343。高分辨質譜(HRMS)計算值C11H4BrF6NNa(M+Na)+:365.9329,實測值:365.9346。
